The hard fungal fruit body especially the Xylariaceae fungi are made up of hard tissues which are commonly known as dead men's finger. The fruit bodies are very difficult to make a good thin section for their microscopic anatomical studies, due to their hardness or very fragile characteristics. Also, temporary slides are not efficient to store for a longer time as well as mycologists face a lot of problems regarding the future observation of a particular sample and the lack of proper tools and techniques of fungal histology still a very trackless research area. That is why preparing a permanent slide for relevant scientific observation is still very poor, therefore, this present study trying to establish an alternative method to prepare a permanent slide of hard tissue of fungal fruit bodies by applying Microtome techniques with the suitable fixative agent.
